炭疽杆菌是炭疽病的病原体。炭疽杆菌产生的主要毒力因子是γ连接的聚-D-谷氨酸胶囊和由三种成分组成的外毒素，保护性抗原（PA），致死因子（LF）和水肿因子（EF）。这些蛋白质不是单*毒的，而是以二元组合起作用。PA（细胞结合成分）与LF（酶部分之一）的复合物被称为“致死毒素”并可导致死亡。PA和酶促EF一起引起皮肤水肿。分泌后，PA被膜肽酶切割，使63kDa羧基末端片段寡聚化成七聚体。切割是暴露EF和LF的结合位点的必要步骤。组装的PA复合物和相关的LF或EF通过胞吞作用进入细胞。PA介导LF和EF向细胞质的转移，其中这些酶识别并蛋白水解其靶标。LF组分是一种锌依赖性金属蛋白酶，可裂解丝裂原活化蛋白激酶激酶（MAPKK），从而削弱其功能，从而阻断MAPK信号通路。EF是钙调蛋白依赖性腺苷酸环化酶，其在细胞中产生过量的环AMP，导致水肿。目前的研究集中于开发有效的疫苗以及筛选LF酶活性的有效抑制剂。
含有炭疽LF切割位点的FRET肽底物可作为MAPKKide®获得，并可用于评估LF的酶活性。
炭疽保护性抗原片段（PA 20）
从炭疽芽孢杆菌
保护性抗原（PA）是炭疽毒素的受体结合成分。原始的83kDa蛋白质被细胞弗林蛋白酶样蛋白酶切割，形成活化的63kDa单体PA63。在这种形式中，PA63与其他PA63分子结合形成环状七聚体。PA63还具有对于与细胞表面上形成的孔相邻的酶LF和EF特异性的结合位点，用于易位。对于醉酒细胞，PA应与EF或LF一起使用约7倍过量。
