二、核心能力与技术基础
1. 糖科学(Glycosciences)技术平台
GlycoDiag的核心技术平台围绕凝集素与糖链之间的相互作用展开,旨在识别和比较特定的糖基化特征。其GLYcoPROFILE技术采用标准微孔板形式,无需昂贵的专用设备,适用于高通量筛选,且样品无需纯化即可进行分析,适合工艺优化与过程控制。
基于该技术,公司开发了LEctPROFILE系列试剂盒,用于快速评估粗提或纯化糖复合物与凝集素的相互作用,检测时间通常在3小时以内。该系列涵盖多个靶向产品:用于检测末端半乳糖(Galili表位)的Galili LEctPROFILE® kit、用于研究GalNAc糖链结构的ASGPR LEctPROFILE® kit、用于甘露糖化糖链结构检测的DC-SIGN & Langerin LEctPROFILE® kit及FimH LEctPROFILE® kit、用于β-半乳糖苷结构研究的Galectin 3 LEctPROFILE® kit,以及用于细胞表面糖基化分析的Cells LEctPROFILE® kit等。此外,Glycoprotein LEctPROFILE® kit支持在24种凝集素上对糖蛋白进行糖基化水平的系统分析,可用于糖蛋白批次间一致性监测。

2. 微生物群(Microbiota)研究体系
在微生物群领域,GlycoDiag建立了基于野生型菌株的体外共培养模型,这些菌株分离自健康志愿者的特定部位(如面部),相较于标准实验室菌株更能反映自然行为。共培养模型通常由4至5种代表特定皮肤微环境的细菌和/或酵母组成,在同一条件下共同生长。
目前覆盖的皮肤微生物生态系统包括:干性、湿性、油性皮肤、头皮、面部、口腔等。该模型已用于评估化妆品成分对微生物群的影响,结果可区分为杀菌、益生元或维持菌群平衡(“微生物群友好")等作用。其部分研究成果已通过同行评审发表。
三、解决实验中的哪些问题?
1. 糖基化分析与质控问题:在生物治疗药物研发中,早期检测非预期的糖基化是质控的重要环节。LEctPROFILE试剂盒可在工艺开发阶段提供糖基化特征的快速反馈,无需复杂的样品前处理即可完成。
2. 糖蛋白表征与批次一致性:重组糖蛋白生产过程中,批次间的糖型差异可能影响疗效与安全性。Glycoprotein LEctPROFILE® kit能够在24种凝集素层面提供糖基化模式的可视化比对,辅助监测工艺稳定性。
3. 微生物群影响评估的模型问题:化妆品及营养品行业在验证原料对皮肤或肠道微生物群的影响时,常受限于缺乏生理相关的体外模型。GlycoDiag基于野生型菌株的共培养模型模拟了特定微环境的温度、pH及菌群比例,为评估产品是否“微生物群友好"提供了实验依据。
4. 候选分子筛选:LEctPROFILE试剂盒的竞争模式可用于筛选特定凝集素的拮抗剂,评估候选分子的半抑制浓度(IC50)或亲和力比较,适用于药物发现阶段的早期筛选。
四、常见问题与解答
问:试剂盒是否只能用于纯化样品?
答:不是。LEctPROFILE试剂盒可直接用于粗提样品或未纯化的混合物分析,无需对样品进行预先纯化,这简化了工艺优化过程中的样品处理流程。
问:使用这些产品是否需要专门的糖生物学知识或设备?
答:GLYcoPROFILE技术在设计上考虑了非专业人士的使用需求,基于标准微孔板平台,可利用常规实验室的酶标仪进行读数,无需购置专用设备。试剂盒提供标准化的操作流程和已验证的试剂,便于在自有实验室中转移实施。
问:共培养模型使用的是标准菌株还是更具代表性的菌株?
答:GlycoDiag优先使用从健康志愿者特定部位(如面部、头皮)分离的野生型菌株,而非仅依赖标准实验室菌株。这些野生型菌株保留了与原始微环境相关的自然行为特征,在模拟生理条件下可能更具代表性。
问:这些产品和服务是否适用于临床诊断?
答:不适用。LEctPROFILE试剂盒及相关产品明确标示为“仅限研究使用"(For research use only),不适用于临床诊断目的。
